Twelve weeks after immunization, spleen lymphocytes isolated from different groups of mice were stimulated with 5 μg/mL of TsoAg or 1 μg/mL of Con A for 6 h at 37 °C. Total cellular RNA was then extracted for the assay of IFN-γ production by real-time PCR.

The fold change was determined by dividing the quantity of specific IFN-γ mRNA from stimulated cells by the quantity of IFN-γ mRNA from the unstimulated cells.
